The Dempsey Challenge features 10, 25, 50 and 100-mile cycle route options and a 5-kilometer (3.1 mile) and 10-kilometer (6.2 mile) course for walkers and runners.

All routes start and end at Simard-Payne Police Memorial Park near the Androscoggin River in Lewiston.

All cycle routes cross the Androscoggin River and loop in a westerly direction, circling back at different points. The 100-mile ride extends through the rural towns of Poland, Raymond, Naples, Bridgton, Harrison, Casco, and Otisfield, then back through Poland and Auburn to Lewiston.

The 5K run/walk meanders through downtown Lewiston, over the Bernard Lown Peace Bridge to Auburn where is follows Main Street to the the Riverwalk, then along the Riverwalk from Auburn to Lewiston via a pedestrian-only bridge.
